How should I answer the question, what are three positive characteristics you don't possess?

You are here to get selected for the job profile. So, it would be better to not give out the primary skills required for the job as your area of weakness.

For e.g. A candidate applying for business development can’t afford to say that he is bad at communication. This would be like throwing your own wicket.

If you think you are missing out on any primary quality required for the type of career you aspire for, it’s better to take out some time and train yourself.

However, it is impossible for a person to not have any weakness. So, list out certain things that you lack while keeping a focus on what are you doing to improve yourself.
